What is Janoshik reporting in mg

I’m so confused now because I was readng last night and Peter Magic said the mass they report on the typical peptide analysis is not the net peptide content, because we don’t need it.

He also said the purity only means the percent of the desired peptide among the peptide/fragments, NOT what %of the vial contents is your peptide, how do we know how much medication is actually present? (The problem is that lyophilized peptide has the counterion, so if we weigh the puck, it’s not all med. The counterion and residual solvent is not identifiable using the hplc method he uses, he said.)

Related, I’m also now unsure whether or not fillers are in the puck, for tirz or other peptides. I have read that everyone puts it in and also that they would be identified inpurity tests, that the fillers stabilize the lyophilate and make the medication even visible, but is any of that true?

Partly answering my own question piece by piece lol.

Yes there must be filler in the vial bc 10mg of product would be barely visible. Knowing that, mannitol and other sugars are used to stabilize the delicate peptides while undergoing the lyophilization.

Also knowing that, I’d guess that once I reconstitute, freezing tirz again is likely to be safe enough (freeze in the coldest part of your freezer, like where the air blows from) since it had to undergo freezing already. That’s what I’m going to go with, that plus others report success. And supposedly testing has been done (I have not gotten into the PTDS group yet, but supposedly there is something there, I hear. If anyone knows of that…)
 
Why recon lyophilized powder only to freeze it again? Seems counter intuitive especially if more than 1 vial at a time.

That’s a great idea also. I can see it really would be a bit of effort to set up a clean work area and do all the mixing and filtering. Not that I don’t love playing with my hoard over detailed labwork, but my cat can at any time get let into the room by my family and cause trouble.

Just seems silly to me to freeze a bunch of vials and make the subject to temperature degradation if the power or freezer fail vs shelf stable non-temp sensitive material. I just can't wrap my head around the risks of having to trash hundreds of dollars of product over a 5 minute process, but to each his own. 🤷‍♂️

Not that I’m trying to convince anyone else, but I’d actually been thinking that specific risk is likely far less than it seems at first glance. Tirz itself is stable weeks to possibly months (according to some hearsay from compounders). So if you freeze inside a thermos or thermal mass to modulate temperature swings, you probably don’t have to worry unless the power goes out for a month. And after a day you’ll probably know and have done something. And the freeze thaw seems to cause destabilization on the order of 1% (I’ve heard it in those janoshik interviews that it’s 0.2% for lyophilization, which involves freezing… using a stabilizer… which is still in your recom from the puck btw…)

That, and also I am contemplating options, I just got a number if those ProRx vials, someone called it “toilet tirz” this morning. They were $$$ now I don’t feel so great about them. Powders generally cost less in larger mg vials. I wish ignorance is bliss weren’t so true.
 
Someone might have said that if they sell already reconstituted Tirz. That's because you really have no idea what's in it, both the peptide and the BAC they used and there is almost never a COA. It's also the most expensive way to go, but we already chatted about that. If you really want a US seller, use Nexaph. They sell 10 vial kits of Reta, Tirz, etc., unreconstituted and with a Q Code on the box that links to the Jano COA, so at least you have some idea. It's expensive unless you get a pre-sale but those happen regularly. Then, you can just reconstitute (with Hospira, hopefully) as you go without this freezing business.

I actually was thinking I don’t want another layer of dubiously trustworthy middlemen to both add deliberate shortcuts and inadvertent errors. I will have to suck it up and figure out bitcoin’s intricacies. (I know that sounds harsh but wow the ProRx 483 illustrates what can go badly in the US too. I also have an idea how hard it is to train people to be meticulous and accurate. And then if you get someone awesome they move on so they can be you 🤣, which is awesome but it means that staffing is always variably skilled.)

I’m on all these forums and discords suddenly and I can’t keep them straight so I can’t go back and find where that came from so take it with the usual grain of salt.
I spent a couple hours tonight setting up a workspace to filter my 288mg in 16ml of liquid tirz of dubiously trustworthy BUD.

The Reta I got was 99.887%. The COA is tied right to the box.Best of luck.

That's what I'm using right now. Down 32 lbs in 11 weeks. Eliminated the urge for alcohol, full appetite suppression and I feel great. It's amazing stuff.

I have some of the "toilet triz" too! Hadn't heard it called that yet, but totally get it. I'm not feeling the love for those vials either (5 of them). My plan is to hold off using the vials and see what happens in the subreddit. And then, I'm thinking I might filter prior to use.
