Had a bit of a rough experience with PT-141 yesterday and I wanted to share in case it helps anyone dialing in their dose.

I'd tried it twice before at conservative amounts: 0.5mg first (felt nothing), then 1mg (still nothing). So yesterday I decided to bump up to 2.5mg. Turns out, that was too much. About 45 minutes in, it definitely kicked in. After... taking care of business... the erection simply didn't go away. We're talking roughly 6 hours , maybe more since I fell asleep (with a boner!). On top of that, my appetite completely vanished, and this morning I woke up feeling like I'd run two marathons: full-body aches, the works.

Lesson learned: go in half gram increments 😛

Well, I think you should read the linked post, and the related article, I linked it below for your convenience. The titles are missing the interesting part: "the last erection you may ever have". I hope you dogged the bullet, it's not the same substance, but you never know, mechanism can be the same. Be careful next time!

Andrew Huberman Raises Risk On Tanning Peptide: It Makes People Tan From The Inside, Raises Energy And Libido Dramatically, Fat Loss Dramatically

Neuroscientist Andrew Huberman appeared on the a16z podcast to discuss peptides, sleep technology, and the future of obesity treatment. During the conversation, he talked about peptides and specifically, melanotan. He first introduced it while listing peptides that have actual human data behind...

Are all y'all seriously screwing around with PT-141 without first reading up on priapism and making note that if it's been four hours, you need to be on your way to the ER?

I did read up on it, and I was aware of the 4 hour ER recommendation. This is not the first "male enhancement" substance I have tried, so the prolonged erection was not new to me. It's working just fine today.

That's cool, you do what you want, I just don't want other guys who are a little more risk averse when it comes to their Don Johnson reading "six hour boner, fell asleep, lulz what a trip" and thinking that's not, like, a medical emergency.

LMAO fair enough.

Overnight erections from PT-141 are not usually problematic like true, ischemic priapism. The biggest danger is taking other meds along with PT-141, like trazodone or antipsychotics:

PT-141 experience

Mid 40's male, no problems or issues with sexual function, but I was interested in what PT-141 might "feel" like so dosed 1.75MG and injected IC last night, about 3 hours before bedtime, about 1 hour after dinner. I felt absolutely nothing. I listened to some music for 3 hours, then fell asleep...

I won't take more than 1 mg of PT-141 in 24 hours (again).

The number of hours can help with diagnosis but may not be as important as other factors/symptoms:

Yeah, when I tried Viagra 2 decades ago, that erection HURT. This erection was more of a nuisance.

Lower-limb exercises like squats or using an elliptical machine may help with priapism:

Conservative management strategies for ischemic priapism are sparsely described in the literature but generally include ice pack application to the area, cold showers, masturbation and rarely, exercise. These strategies lack sound evidence, but the risks of attempting them are minimal as long as access to more definitive treatment is not delayed. Lower-limb exercise as a first-line treatment warrants further study in the undifferentiated emergency department priapism population. The case we present and discuss here illustrates the potential benefits of a trial of acute lower-limb exercise, specifically stair climbing, as a treatment for medication-induced priapism.

Huberman was referencing melanotan. PT-141 (bremelanotide, brand name Vyleesi) is FDA-approved.

Trying to figure out how to square these two. Are full-body aches a side effect just from taking it? I'd always assumed they were a result of working your body so hard that every muscle hurts, but perhaps that's a fault assumption. Or perhaps you have a form of being your own partner that's a full body workout?

This went from a general post for others to benefit from my negative experience and to help others make better decisions when experimenting with PT-141 to a TMI information session 😛

I was NOT expecting the extreme muscle soreness in the morning so I did a bit of investigation and apparently this is a known, but not often experienced side effect. One resource even described it as "flu like muscle aches" which is exactly how I would describe it.

I'll note that all of the soreness, and the constant feeling of needing to stretch the muscles, has gone away after a few hours.
