Update week 2. Took 2nd dose yesterday. 1 mg again. As you can see with the 14-day 1/2 life, the medicine has increased. The level of saturation will only increase. The half-life is twice the weekly dosage. No other GLP has this long a half-life. I have tried to time this 2 days prior to my normal weekly Tirz (15mg) shot. Again this week, those days the food suppression, which normally fades, has been suppressed. I am eating because I have to hit my Calories. Yesterday, I noticed an 8-point uptake in RHR. That has subsided. The thirst and dry mouth remain but have reduced. All is good. If stacking low and slow is the move. Down 3 lbs in the last 10 days. At this stage (13 months), that number is well above average, as my weight loss has come to a crawl.

I'm just getting started, at a dose of only 0.5mg. I'm going to increase it to 1 mg next dose. At 0.5mg I'm not noticing any particular side effects, and any hunger suppression is subtle. I guess I need to be less timid in my dosing.

I'm trying out the Regimen app, so the half-life calculation will be helpful.

Week 5 update. I have decided to titrate up to 2mg.

After all, I am just a monkey in training. Good suppression again and mid elevated fatigue that lasted 1 afternoon. Did sleep very well. Just blasted my 15mg of Tirz and the party starts again.

Really do enjoy this combo. Will keep an eye on the fatigue. It was similar to the fatigue I felt with Tirz in the beginning, which now has subsided. As you can see, at 2 mg, the medication level has nearly doubled due to its 14-day half-life.

According to the phase 1 studies on eloralintide, not only elimination, but also resorption of eloralintide is slow. The time to peak concentration is 3–5.5 days. Whoever made the calculations for you curve didn't take that into account. Your curve suggests it's closer to 1 day, if that. So it doesn't give a realistic picture of the levels you are at. The peaks are too steep and too high.

Eloralintide demonstrated a distinct PK profile in the phase 1 trial. It was absorbed with a median tmax of 72–132 h across the doses ranging from 0.04 to 12 mg. Moreover, it had a half-life of 310–366 h (12.9–15.3 days) across doses.

The curve is a bit jagged because it has not been smoothed. But I think you can see it looks different, especially in the beginning.
