Orforglipron: The next big thing?

Update: test subject is still seeing a slow but steady weight loss, along with mild side effects.

Switched to 12mg pills, because liquid is still horrible.

This shit works, but behaves like Sema. Go too fast without experience, and you’ll regret it.

Are people taking these by myself or along with an injectable glp? I have some on hand but haven’t decided what to do with them

Both. Orfo is the only GLP on the (grey) market that is potentially ideal for stacking GLP-1s. Since it is tiny compared to the other GLPs, it can fit onto the same GLP-1 receptor as sema/tirz/reta.

I think the easiest way to start is to take at the end of a tirz/reta/sema week. And see how you feel the next day. That is basically what I did. But I wouldn't take something like 12 mg. Would use a pill splitter for such higher-dose tablets when starting. (Newbies with no GLP experience should not take more than 3 mg to start, ideally 1 mg as in the last trial.)

Thanks. You think 1/4 of a 12 would be ok?

Yes. I started with 9.3 mg (tested amount of 8 mg Uther tablet), which is what I still take. But the sides were definitely there by starting at that dose.

Is there no coating or something on the tablet? I am aksing because, splitting would destroy special coatings.

Uther says not to split. But no issues when trying it myself, out of curiosity. I avoid splitting, but it is the lesser of two evils, if starting with 12+ mg tablets.

An alternative to splitting is taking less often, like every other day.

Yeah, thought so. When you split it, the stomach acid-resistant coating is damaged and the medication has little or no effect. That is the disadvantage of these tablets.

That was not the issue for me. The split tablets seemed to work just as well.

Orfo itself is stomach resistant: "Orforglipron is inherently resistant to stomach acid and does not require a protective coating to remain effective" (Gemini). I may find an article later.

Still other concerns though, more in theory than anything else. Like one side of the tablet may be stronger than the other.

Seems like that depends on who's manufacturing the pill. I have no doubt that the above is true if Lilly is the manufacturer. I'm less convinced this is the case for gray vendors.

The third/last phase of the orfo trial used 1 mg as a starting dose. Phase two used mostly 3 mg as the starting dose, with more GI side effects.

But the smallest dose available in grey is 6 mg, and some resellers like PGB only sell 12 mg (tested at 13.7 mg). Uther has 8 mg (tested at about 9 mg) and 12 mg (tested at about 13 mg). WWB has 6 mg but no testing available.

No mass testing on their COA? I see it passed endotoxin testing though.

Though it's good they have the 6 mg, their price per mg is the same as 12 mg elsewhere. And it doesn't take long to titrate up to 12 mg, which is the sweet spot now for orfo tablets.

So if we get 12mg or 6mg tablets it's best to cut in 4ths or 8ths for testing and research to start off with?

There are people who think splitting a tablet is somehow crazy. But I would split when necessary and have before. I am too cheap to pay for 6 mg tablets.

I researched a bit about the tablets. There should be a special coating on it. So splitting makes the medication less effective. But I dont know how they manufactor it in china.

So get ready for a wild ride on the porcelain God if you just start at 12/6mg? #gotit.

My guess is that 3 mg is a safe starting dose for those who have been on GLPs before. 3 mg was mostly used as the starting dose in the second phase of the orfo trials. But only 1 mg was used as the starting dose in the third phase/study.

The Uther tablets are small but can be split into four pieces. I used a pill splitter for spitting in two. But to split again, I used my fingers instead. The smallest piece can be used for the first dose.

Another option is to take every other day or so. We will know more when more people start reporting their experiences.

I started at 8 mg of Uther, tested at 9.3 mg. GI sides were definitely moderate/strong at times, even though I was mostly on a break from tirz.

I skipped a day after day three, since the sides were getting a little tiring. Only after that did I try splitting the tablets, but I really didn't need to at that point.

Right now, I am on the 12-mg Uther tablets daily (tested at about 13 mg), along with a low dose of tirz (which I will be raising again).

Unlike the ritual of a weekly injectable, it's much easier to forget whether you've already taken a pill. So to avoid accidentally overdosing on orfo (like taking two pills in one day), I would use a weekly pill reminder case.

Personally, I am done stacking with orfo. I will just save it for a maintenance phase on its own. Had some bad GI sides, especially bloating, last night.
